Gamma Radiation and Tissue Culture Induced Variability in Moricandia (Moricandia sinaica Boiss) with Genetic Assessment by ISSR markers and Phytochemical Profiling of Organosulfur Compounds via GC–MS

Abstract

Moricandia sinaica is a member of genus Moricandia. Moricandia sinaica is used in traditional medicine to alleviate certain disorders such as syphilis and exhibits analgesic, anti-inflammatory, antipyretic, antioxidant, and antigenotoxic proper- ties. Buds were cultured on Murashige and Skoog (MS) solid medium with dif- ferent strength with and without hormones asBAP and Kin. The best medium was half strength MS without hormones. Plantlets were exposed to different gamma radiation doses 10, 20, 30, 40, 50 and 60 Gy.The morphological param- eters of irradiated plantlets were decreased by increasing gamma radiation doses. The lethal dose was 40 Gy and the optimal dose for explant survival was 10 Gy. Genetic diversity was evaluated by ISSR markers using 20 primers. 149 bands were the total amplified bands by ISSR markers. The mean of polymorphism was 58.55%.The polymorphic information content (PIC) with an average 0.363 (PIC<0.5) classifying most primers as moderately informative. High PIC values (0.371–0.374)and discriminating power confirmed substantial genomic varia-
tion induced by Gamma radiation treatments. The effect of gamma radiation on organosulfur compounds as analyzed by GC- MS was limited to increased production as2-Myristynoyl pantetheine; reduced production as Ethanimidothioic acid,2 (Dimethylamino)N[(Methylamino)Carbonyl]Oxy]-2-Oxo-,Methyl Ester; disappearance of some compounds as Thiocyanic Acid, Octyl Ester and 2-[3-Cyclohexylaminopropylamino] ethylthiophosphate and formation of some new compounds as Leukotriene D4 methyl ester, Cefazolin, Meraptal, Pentaacetate, Mannitol, tert-Hexadecanethiol, 2-Ethyl-5,7-Dimethyl.
